逍遥散对慢性束缚应激肝郁脾虚证模型大鼠杏仁核Bax和Bcl-2蛋白表达的影响

Effect of Xiaoyao Powder on the expression of Bax and Bcl-2 in amygadala of rats induced by chronic restrain stress

目的:研究逍遥散对肝郁脾虚证模型大鼠杏仁核Bax和Bcl-2蛋白表达的影响。方法:36只SD雄性大鼠随机等分为正常组、模型组和逍遥散组。以14d慢性束缚应激造肝郁脾虚证模型组,除逍遥散组灌胃逍遥散悬液外,其余组均给予等量0.9%氯化钠溶液。取材后,通过Western Blotting方法观察各组杏仁核Bax和Bcl-2蛋白表达。结果:与正常组比较,模型组杏仁核中促凋亡因子Bax明显增多,而逍遥散能相应改善此种趋势;抑凋亡因子Bcl-2在3组杏仁核中表达无统计学差异。结论:逍遥散能够通过降低Bax的表达来改善慢性束缚应激致肝郁脾虚证模型大鼠杏仁核神经细胞的凋亡。

Objective： To investigate the effect of Xiaoyao Powder on expression of the apoptosis factor bax and Bcl-2 in amygdala,which induced by the chronic restraint stress in the rats.Methods： 36 male SD rats were randomly divided into 3 equal groups： control,stress,Xiaoyao Powder.After 14 days restraint stress,the protein expression of Bax and Bcl-2 in amygdala in these 3 groups were examined by west blotting analysis.Results： Compared with control group,expression of bax in amygdala in stress group was increased significantly.However,Xiaoyao Powder could reverse this potential,as compared with the stress group,and the differences were significant.What＇s more,there were no significant differences among these three groups on the expression of Bcl-2 in amygdala.Conclusion： Xiaoyao Powder may effectively improve the apoptosis situation of amygdala,which induced by chronic restraint stress by increasing the protein expression of bax.
