参芪复方对GK大鼠2型糖尿病大血管病变氧化应激的影响

Effects of Shenqi Compound Recipe on oxidative stress in GK rats with typeⅡ diabetes mellitus macrovascular complications

目的:观察参芪复方对GK大鼠2型糖尿病大血管病变氧化应激的影响。方法:采用在GK大鼠饮水中添加Nω-硝基-L-精氨酸甲酯(L-NAME)0.1mg.mL-1.d-1联合高脂饲料喂饲的方法复制2型糖尿病大血管病变模型。GK大鼠随机分成GK空白组、模型组、阿托伐他丁组、参芪复方组,另设Wistar大鼠为正常对照组。造模同时给予相应药物,周期35d。实验结束后测定大鼠血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性水平;RT-PCR法测定主动脉还原型辅酶Ⅱ(NADPH)氧化酶亚基p22phox mRNA及p47phox mRNA表达。结果:参芪复方组GK大鼠血清SOD、GSH-Px活性较模型组升高(P<0.01,P<0.05),阿托伐他汀和参芪复方均能降低主动脉p47phox mRNA、p22phox mRNA的表达水平(P<0.05)。结论:参芪复方可提高糖尿病GK大鼠血清SOD和GSH-Px活性,下调主动脉p47phox mRNA、p22phox mRNA表达,可能因此减轻糖尿病大血管病变机体的氧化应激水平,这可能是参芪复方治疗T2DM大血管病变的机制之一。

Objective： To study the effect of Shenqi Compoumd Recipe（SQCR） on the oxidative stress in GK rats（Goto-Kakizaki wistar rats） with T2DM Macroangiopathy.Methods： Copied T2DM model with macrovascular complications by Nω-nitro-L-arginine methyl ester（L-NAME） 0.1mg·mL-1·d-1 added into the drinking water plus high fat diet.GK rats were divided into GK group,model group,atorvastatin group,SQCR group and another 14 Wistar rats act as the normal group.Treating while setting up modelfor 35 days.Antioxidant enzymes was examined with colorimetry while the mRNA expression of p22phox and p47phox in thoracic aorta vessel wall by real-time PCR.Results： The SQCR group had significantly higher GSH-Px activity compared with model group,and higher activity of both SOD and GSH-Px compared with atorvastatin group.Compared with model group,the mRNA expression of p47phox and p22phox decreased in atorvastatin group and SQCR group.Conclusion： Improving the activities of antioxidant enzymes（including SOD and GSH-Px）,signi？cantly downregulating the mRNA expression of p47phox and p22phox in aorta in GK rats with T2DM Macroangiopathy maybe one of the possible curing mechanisms that inhibit the oxidative stress and prevented T2DM Macroangiopathy by ShenQi compoumd recipe.
