摘要
目的:探讨尿毒症腹膜透析大鼠腹膜组织中缺氧诱导因子1α(HIF-1α)和结缔组织生长因子(CTGF)的表达及意义。方法:32只雄性SD大鼠随机分为正常对照组、假手术组、尿毒症组及腹膜透析组,每组8只。尿毒症和腹膜透析组大鼠制备尿毒症模型,腹膜透析组于尿毒症造模成功后第6周开始每天规律行腹膜透析,28d后取4组大鼠新鲜腹膜组织,采用免疫组化及RT-PCR技术检测HIF-1α、CTGF蛋白及mRNA的表达水平,并采用免疫组化技术检测α-平滑肌肌动蛋白(α-SMA)的表达。结果:4组大鼠腹膜组织中HIF-1α、CTGF蛋白和mRNA的表达水平及α-SMA蛋白表达水平差异均有统计学意义(P均<0.001),尿毒症组上述指标较正常对照组升高,腹膜透析组较尿毒症组升高更明显(P<0.05)。4组腹膜组织中3个蛋白的表达均呈正相关(P<0.05)。结论:腹膜透析大鼠腹膜纤维化的发生可能与HIF-1α、CTGF的表达上调有关。
Aim:To investigate the expressions of hypoxia-inducible factor-1α(HIF-1α) and connective tissue growth factor(CTGF) in peritoneum of peritoneal dialysis(PD) rats.Methods:Thirty-two male SD rats were randomly divided into four groups:normal control group,sham operation group,model group and PD group.Rats in model group and PD group were to make renal failure model.Six weeks after the model being successfully made,regular peritoneal dialysis in PD group was performed for 28 days.Then the protein and mRNA expressions of HIF-1α and CTGF were detected in peritoneal tissue of the 4 groups by the immunohistochemical staining and RT-PCR.The expression of alpha-smooth muscle actin(α-SMA) was detected.Results:There were significant differences in the expressions of HIF-1α protein and mRNA,CTGF protein and mRNA,and α-SMA protein among the 4 group(P0.001).Compared with those of the control group,the indexes mentioned above were higher in model group and PD group(P0.05),and the indexes mentioned above were higher in PD group than those in model group(P0.05).The expressions of HIF-1α,CTGF and α-SMA protein in the 4 groups were positively related to each other(P0.05).Conclusion:Peritoneal fibrosis in PD rats may be related to the upregulation of HIF-1α and CTGF expressions in peritoneum.
