重组人p53腺病毒注射液联合顺铂对宫颈癌HeLa细胞增殖及VEGF表达的影响 被引量:2

摘要 目的 评估重组人p53腺病毒注射液(rAd-p53)联合顺铂(DDP)对宫颈癌HeLa细胞的增殖及血管内皮生长因子(VEGF)表达的影响.方法 采用四甲基偶氮唑蓝法(MTT)测定不同浓度rAd-p53、DDP、rAd-p53联合DDP对HeLa细胞增殖的影响;采用Western blot法测定rAd-p53、DDP、以及两药联合应用对HeLa细胞中VEGF表达情况的影响.结果 rAd-p53、DDP、rAd-p53联合DDP作用于HeLa 细胞时,细胞增殖均被抑制,两药联合组细胞增殖抑制率明显高于两药单用组.rAd-p53、DDP单药作用及两药联合作用48h后HeLa细胞中VEGF表达均下调,且联合用药组较单药组抑制明显.结论 rAd-p53和DDP均能抑制HeLa细胞的生长,rAd-p53联合DDP比两药单用对HeLa细胞的生长抑制作用更显著;rAd-p53可能通过下调VEGF活性抑制宫颈癌HeLa细胞增殖,抑制肿瘤血管生长,增强DDP的作用效果. Objecttive To evaluate the efficacy of recombinant adenovirus - p53 ( rAd - p53) combined with cisplatin( DDP)on the cell proliferation and vascular endothelial growth factor (VEGF) expression in human cervical HeLa cells. Methods After HeLa cells were treated with rAd - p53, DDP and the combination of rAd - p53 and DDP, re- spectively, the staining inhibitory effect of them on HeLa cells growth was analyzed by MTT assay, Western blot was per- formed for the changes in expressions of VEGF. Results The proliferation of HeLa cells was significantly inhibited after being treated with rAd - p53 or DDP or combination. This inhibitory effect was significantly enhanced by the combination group. The expression levels of VEGF decreased in HeLa cells waa significantly decreased in control group compared with rAd -p53, DDP and combination for 48h, and in the combination group, the inhibiting expression effect iwas more im- pressive. Conclusion rAd - p53 or DDP can inhibit the proliferation of HeLa cells, but rAd - p53 combined with DDP can enhance the inhibitory effect on proliferation of HeLa cells, rAd - p53 maybe inhibit the proliferation of HeLa cells through down - regulating the activation of VEGF and thus inhibit the growth of tumor vessel, rAd - p53 combined with DDP can generate synergistic effect.
