摘要
目的观察乌头碱对H2O2构建的心衰细胞模型的强心作用的量-时-效关系。方法利用原代培养的乳鼠心肌细胞,加H2O2构建心衰模型,采用均匀设计,分别加5、10、25、50、100μmol/L的乌头碱作用0.5、1、2、4 h及8 h后测MTT,然后进行验证实验。结果 均匀设计预测的最佳组合为乌头碱50μmol/L作用0.5 h,验证实验也证实如此。结论乌头碱对心衰模型有明显强心作用,其最佳量-时-效组合为50μmol/L的乌头碱作用与心衰模型0.5h。
Objective:To observe the dose-time-effect relationship in cardiac effect of heart failure cells induced by H20〉 Methods:These heart failure models, establised by H202 and myocardial cells of neonatal rats, were ana- lyzed by the uniform design method. These models were divided into several groups which were used respectively by the dose of 5 Ixmol/L, 10μmol/L,25μmol/L,50μmol/L and 100μmol/L for 0.5 h, 1 h,2 h,4 h and 8 h sepa rately. Results:The best predicted combination of uniform design was the 50μmol/L of aconitine effect for 0.5 h. Meanwhile,the validation experiments also confirmed that. Conclusion:The cardiac effect by aconitine on a cell model of heart failure is significant. The best combination of dose-time is 50μmol / L and 0.5 h.
