摘要
目的探讨上调缺氧诱导因子-1(HIF-1)对大鼠球囊损伤后颈动脉舒张反应的影响。方法 30只雄性健康Sprague Dawley大鼠随机分为假手术组、模型组和L-Mim组。模型组和L-Mim组均采用1.5F球囊导管建立颈动脉球囊损伤模型,于术后第1 d开始,L-Mim组给予含羞草素100 mg/(kg.d)灌胃,假手术组和模型组给予等量生理盐水代替。干预14 d后,取颈动脉损伤段,检测乙酰胆碱和硝酸甘油诱导的血管舒张反应。结果与假手术组比较,模型组血管对乙酰胆碱的舒张反应显著受损,最大舒张反应显著降低(P<0.01),而L-Mim组最大舒张反应较模型组显著增加(P<0.01)。3组血管对硝酸甘油的舒张反应无显著差异。结论采用L-Mim上调HIF-1可部分对抗球囊损伤对血管内皮功能的损害。
Objective To investigate the effects of up-regulating hypoxia inducible factor-1(HIF-1)on dilatation of carotid arteries after saccule injury in rats.Methods Thirty male healthy SD rats were randomly divided into sham operation group,model group and L-Mim group.Model of carotid arterial saccule injury was established by 1.5 F Foley's tube in the model group and the L-Mim group.From the first day after the operation,the L-Mim group received the intragastric administration with 100 mg/(kg·d) leucaenine;the sham operation group and the model group received the intragasgtric administration with the same dosage of saline.After the intervention for 14 d,the injured segment of carotid artery was obtained for the detection of acetylcholine-and nitroglycerin-induced vasodilatations.Results Compared with the sham group,the acetylcholine-induced vasodilatation was significantly impaired in the model group,and the maximal dilatation decreased very significantly(P〈0.01).Moreover,the maximal dilatation increased very significantly in the L-Mim group compared with that in the model group(P〈0.01).There was no significant difference in nitroglycerin-induced vasodilatation among the three groups.Conclusion Up-regulation of HIF-1 with L-Mim may partly attenuate the saccule-injury-induced impairment of blood vessel endothelium function.
