姜黄素对体外培养的人食管癌Ec-109细胞增殖和凋亡的影响 被引量:2

摘要 目的研究姜黄素对体外培养的人食管癌(esophageal carcinoma 109,Ec-109)细胞增殖和凋亡的影响。方法采用噻唑兰(methyl thiazolyl tetrazolium,MTT)法检测不同浓度姜黄素对人食管癌Ec-109细胞增殖的影响;流式细胞术检测Ec-109细胞的凋亡情况;免疫细胞化学方法检测Fas、增殖细胞核抗原(proliferating cellnuclear antigen,PCNA)蛋白表达的情况。结果姜黄素可显著抑制Ec-109细胞的增殖,呈剂量和时间依赖性,半数抑制浓度(inhibitory concentraltion 50,IC_(50))为22.09μmol/L(姜黄素浓度为80μmol/L作用60h)。不同浓度姜黄素均可诱导Ec-109细胞凋亡。姜黄素可显著下调Ec-109细胞PCNA蛋白表达,显著上调Fas蛋白表达。结论姜黄素可抑制体外培养的人食管癌Ec-109细胞的增殖,诱导其凋亡。 Objective To observe the effects of curcumin on the proliferation and apoptosis of human esophageal carcinoma 109 ( Ec - 109 ) cell line in vitro. Methods The inhibitory effect of curcumin on the proliferation of human Ec - 109 cells at different concentrations was determined by methyl thiazolyl tetrazolium (MTY)assay. The changes of cell apoptosis rate of tumo:r cells were determined by flow cytometry (FCM). The expressions of Fas and proliferating cell nuclear antigen(PCNA) proteins were examined by immunocytochemical technique. Results Curcumin significantly inhibited the proliferation of Ec -109 cells in a dose -and time -dependent manner,with the inhibitory concentrahion 50( IC50 ) of 22.09μmol/L( Curcumin concentration for 80μmoL/L role of 60h). Different concentrations of curcumin can induce apoptosis of Ec - 109 cell. Curcumin dramatically inhibited protein expression of PCNA, while upregulated the expression of Fas protein. Conclusion Curcumin can inhibit the proliferation of Ec - 109 cells and induce the apoptosis of Ee - 109 cells in vitro.
