摘要
目的从黏细菌发酵产物中寻找抗乳腺癌活性物质。方法采用分析型高效液相色谱和制备型高效液相色谱对黏细菌93431菌株发酵产物的甲醇浸提液进行分析与分离,采用MTT法,以人乳腺癌细胞株MCF-7、人胚肺成纤维细胞株MRC作为实验模型,对提取的各组分活性进行测定。结果从黏细菌93431菌株的发酵产物中分离得到4种组分,其中93431-1在质量浓度100μg/mL、作用24 h时,对MCF-7细胞的抑制率达51.65%,对MRC细胞的抑制率为17.54%。结论黏细菌93431菌株中93431-1对MCF-7细胞有良好的抑制作用,且对MRC细胞毒性较小。
Objective To obtain anti-breast cancer substances from the fermentation product of myxobacteria. Meth- ods The crude extracts was analyzed and isolated in methanol from the fermentation product of myxobacteria 93431 using analytical HPLC and preparative liquid chromatography. The toxieities of these components to human breast cancer cells MCF-7 and cells MRC were detected through the MTr. Results Four components were isolated from the fermentation products of myxobacteria 93431, in which the component labelled 93431-I showed the strongest cytotoxicity against MCF-7 cells with the inhibitory rate as high as 51.65% at the concentration of 100 μg/mL, was treated for 24 h. At the same ex- perimental condition, it showed quite weak cytotoxicity against cell MRC with the inhibitory rate of 17.54%. Conclusion Component 93431-I isolated from the myxobacteria 93431 shows high cytotoxicity against human breast cancer cell MCF-7 and weak cytotoxicity against MRC.
