摘要
研究重组磷脂酰丝氨酸合成酶表达条件及其用于酶法合成磷脂酰丝氨酸的反应条件。结果表明,在50 mL培养基中,菌体浓度为OD600=0.6时,用0.5 mmol/L的IPTG,在温度为20℃,转速为120 r/min的条件下,诱导12 h后,重组磷脂酰丝氨酸合成酶活力可达1.33 U/mL,是优化前的1.66倍。酶法转化磷脂酰丝氨酸的最适温度为38℃,最适pH为8.0,最适离子是12 mmol/L Mn2+,最适表面活性剂是1.0%Trition X-100。在最适条件下,磷脂酰丝氨酸转化率可达33.15%。
Study on the optimization of expression of recombinant phosphatidylserine synthase and its reaction conditions.The result showed phosphatidylserine synthase activity was 1.33 U/mL when the cell concentration of OD600 was 0.6 in 50 mL medium inducted by 0.5 mmol/L IPTG and fermented 12 h under 20 ℃ and 120 r/min.This activity was 1.66 times bigger than pre-optimized.The optimum reaction conditions were 38 ℃,pH 8.0,12 mmol/L Mn2+ and 1.0 % Trition X-100.The conversion rate was 33.15 % under the optimum conditions.
