Degradation of Phenol over Immobilized Horseradish Peroxidase on Polystyrene Microporous Membranes

摘要 以聚苯乙烯蜂窝状微孔膜为载体,碳二亚胺为活化剂,将HRP固定在微孔膜上,探讨固定化条件如酶溶液质量浓度、固定化时间对HRP酶活性的影响,以及固定化HRP酶催化氧化苯酚性能.结果显示,固定化HRP适宜酶溶液质量浓度为1 g/L,固定化时间3 h.固定化HRP酶催化氧化苯酚适宜用量为15 g/g,反应时间50 min,H2O2与苯酚的摩尔比为3∶2,在此条件下苯酚的去除率在76%左右.HRP固定化酶可循环使用3次,3次后其对苯酚的去除率降为4.26%,催化性能有待一进步提高. Horseradish peroxidase was immobilized on honeycomb patterned microporous polystyrene membranes with carbodiimide as the activator.The effect of immobilized conditions such as HRP concentration and immobilized time on the enzymatic activity was dicussed.Then the catalytic performances for oxidation of phenol over immobilized horseradish peroxidase were inveatigated.It was indicated that the suitable enzyme solution concentration and immobilized time were 1 g/L and 3 h,respectively,for immobilization of horseradish peroxidase.It was showed from the catalytic oxidation of phenol tests that the suitable immobilized HRP content was 15 g/g,the reaction time was 50 min,and the mole ratio of oxydol to phenol was 3∶2.The phenol removal could amount to around 76 %.It was also indicated that the immobilized HRP could be used 3 times and the phenol removal reduced to 4.26 % after 3 times usage.
