疏肝补肾法对睡眠剥夺大鼠学习记忆及前额叶皮质BDNF蛋白表达的影响 被引量:4

摘要 目的研究疏肝补肾法对睡眠剥夺大鼠学习记忆及前额叶皮质脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)蛋白表达的变化。方法以成年雄性SD大鼠36只,随机分为对照组、模型组、疏肝补肾治疗组。应用168小时睡眠剥夺法复制模型,Y迷宫成绩评定学习记忆能力,以血清超氧化物歧化酶(super oxide dismutase,SOD)水平反映脑组织的损害。取大鼠前额叶皮质冰冻切片,以免疫组织化学法检测BDNF的蛋白表达。结果与对照组比较,模型组的Y迷宫成绩显著下降(P<0.01),血清SOD含量明显减少(P<0.01),前额叶皮质BDNF免疫阳性物明显减少(P<0.01);与模型组比较,疏肝补肾治疗组Y迷宫成绩明显提高(P<0.01),血清SOD含量显著增多(P<0.01),BDNF蛋白表达也显著提高(P<0.01)。结论睡眠剥夺能造成加重脑损伤、学习记忆能力减退以及BDNF表达的减少,而疏肝补肾法能改善学习记忆和脑损伤,可能是通过上调BDNF蛋白的表达以及相关通路蛋白所致。 Objective To study changes of learning and memory and brain-derived neurotrophic factor expression in the prefrontal cortex on rats with sleep deprivation through the method of soothing the liver and tonifying the kidney.Methods36 adult male Spargue-Dawley rats are randomly divided into control group,model group,and treated group.Applying the model of 168 hours sleep deprivation to reflect brain damage with levels of serum SOD and evaluate learning and memory abilities with the results of Y-maze.Using frozen sections in the prefrontal cortex of rats to detect protein expression of BDNF with immuno-histochemical.Results Compared with the control group,Y-maze's results reduced remarkably(P0.01),serum SOD declined(P0.01),and the immune reactive BDNF in prefrontal cortex declined(P0.01) in the model group;compared with the model group,treated groups improved the Y-maze result(P0.01),significantly increased serum SOD content(P0.01),the expression of BDNF protein is also significantly enhanced(P0.01).Conclusion Sleep deprivation can result in learning and memory loss and brain damage,reduction in the expression of BDNF;the method of soothing the liver and kidney can recover learning and memory and brain injury,possibly through increasing BDNF protein expression and relative pathways of protein.
