黄芩苷对金黄色葡萄球菌生物膜抑制作用的体外研究 被引量:27

摘要 目的研究黄芩苷对金黄色葡萄球菌早期、成熟生物膜(BF)的抑制作用。方法以编号为17546金黄色葡萄球菌为试验菌株,构建体外BF模型,分为空白对照组、试验组(黄芩苷组),两倍稀释法测定最低抑菌浓度;荧光显微镜观察载体表面BF形态;连续稀释法行活菌计数。结果建模3d,荧光显微镜发现黄芩苷终浓度分别为1024、512、256μg/ml的试验组形成BF明显少于空白对照组,载体表面活菌计数分别为(5.05±0.27)、(5.17±0.31)、(5.27±0.55)log10CFU/ml,均少于空白对照组(P<0.05);建模7d荧光显微镜观察,黄芩苷终浓度分别为1024、512、256μg/ml的试验组仅见少许散在生物膜,BF结构较空白对照组明显稀薄,其BF内细菌活菌计数分别为(5.19±0.30)、(5.24±0.04)、(5.33±0.22)log10CFU/ml,亦均少于空白对照组(P<0.05)。结论黄芩苷在体外可以抑制金黄色葡萄球菌生物膜的形成。 OBJECTIVE To investigate the inhibitory effect of Baicalin on Staphylococcus aureu biofilm(BF)in vitro.METHODS The in vitro BF model was established by setting S.aureu-17546 isolates as the study strains,which were divided into the control group and Baicalin group;minimum inhibitory concentration(MIC) was measured by using double dilution method;the configuration of the BF on the carries surface was observed by fluorescence microscope;viable bacterial counts were determined by serial dilution.RESULTS On the third day of the building of BF model,fluorescence microscope revealed that the BF formed in the Baicalin group with final concentration of Baicalin of 1024 μg/ml,512 μg/ml and 256 μg/ml was significantly less than those in the control group,the viable bacterial counts of S.aureu-17546 adhered to the surface of carriers in Baicalin group were respectively(5.05±0.27)log10CFU/ml,(5.17±0.31)log10CFU/ml,and(5.27±0.55)log10CFU/ml,significantly less than in the control group(P〈0.05);on the 7th day of the building of BF model,fluorescence microscope revealed that there were a few of scattered BF in the Baicalin group with the final concentration of Baicalin of 1024 μg/ml,512 μg /ml and 256 μg/ml,the construction of BF was significantly rarefied as compared with the control group,the viable bacterial counts inside the BF were respectively(5.19±0.30)log10CFU/ml,(5.24±0.04)log10CFU/ml,and(5.33±0.22)log10CFU/ml,significantly less than(P〈0.05).CONCLUSION Baicalin can inhibit the in vitro formation of S.aureu BF.
