摘要
目的:观察参附汤对心阳虚型慢性心力衰竭大鼠的影响。方法:大鼠随机分为4组,正常组,模型组,地高辛组,参附汤组(按生药量计为5 g·kg-1,ig),除正常组外,均采用大鼠ip阿霉素4 mg·kg-1的方法建立心阳虚型慢性心力衰竭模型,每周1次,共6周。在造模的同时给药组每天ig给药1次,持续7周,末次给药后测心功能,取标本检测血清超氧化物歧化酶(SOD)活性,丙二醛(MDA)含量,心肌钙调神经磷酸酶(CaN)含量、钙含量、活化T细胞核因子3(NFAT3)蛋白的相对表达。结果:与正常组比较,模型组心功能降低,体重降低,心脏指数升高,SOD活性降低,MDA含量升高,心肌CaN含量、钙含量、大鼠心肌组织中NFAT3/β-actin吸光度比值显著升高(P<0.01);与模型组比较,参附汤组心功能升高,体重增加,心脏指数降低,SOD活性升高,MDA含量降低,CaN含量、钙含量、大鼠心肌组织中NFAT3/β-actin吸光度比值降低(P<0.05);参附汤组对受损伤的心肌组织和心阳虚症状有一定的改善作用。结论:参附汤通过抑制CaN-NFAT3信号转导通路治疗心阳虚型慢性心力衰竭。
Objective:To observe the effects of Shenfu Tang on chronic heart failure with heart-yang deficiency.Method: The rats were randomly divided into four groups,normal control group,model group,digoxin group,Shenfu Tang group(5 g·kg-1,ig).The chronic heart failure model with heart-yang deficiency was established by intraperitoneal injection of adriamycin 4 mg·kg-1 once a week,lasted 6 weeks.Meanwhile the corresponding drugs were administrated for 7 weeks.The cardiac function was measured.The serum was collected to examine superoxide dismutase(SOD)activity,malondialdehyde(MDA)content,calcineurin(CaN) and calcium content.The expression of relative phosphorylation and the dephosphorylation protein of nuclear factor of activated T cell 3(NFAT3) was assayed.Result: Compared with control group,model group decreased heart function,body weight and SOD activity,and increased cardiac index,MDA content,cardiac CAN content,calcium content,the light density ratio of dephosphorylation NFAT3/β-actin(P0.01).Compared with model group,Shenfu Tang group increased heart function,body weight and SOD activity and decreased cardiac index,MDA content,cardiac CAN content,calcium content,the light density ratio of dephosphorylation NFAT3/β-actin(P0.05).Conclusion: Shenfu Tang can treat chronic heart failure with heart-yang deficiency by suppressing CaN-NFAT3 signal transduction pathway.
