小鼠C_2早期T细胞株是由内源性逆转录病毒转化的细胞株

C_2细胞是我室建立的又一株小鼠早期T细胞株,此株细胞表现为转化细胞特征,在普通培养基中生长良好,不需外源因子供应。本文报导C_2细胞可使裸鼠致癌,电镜观察可见在C_2细胞浆的特殊基质部位内有大量病毒颗粒,并可见病毒释出于胞外;在从C_2细胞分离出的病毒富集的组份中具有逆转录酶活性,说明C_2细胞中的病毒为感染型病毒。

C2 cell line was established from cultures of mouse spleen cells of Balb/C origen in our lab, with the CD4-CD8- early T cell characteristics. Injection of C2 cells (5×105 cells) into the peritoneal cavity of the Balb/C nu/nu mice, it caused an ascites type of cancer within two or three weeks, and accompanied with the metastasis of tumour cells to the liver, spleen and lymph nodes. Under the electron microscopic examination, a large amount of virus particles was revealed in a special matrix in the cytoplasm near the membrane of the C2 cells, and the virus particles were also visualized outside the membrane.The infectious retrovirus replicated in C2 cells was further confirmed by the high activity of reverse transcriptase present in the virus -enriched fraction extracted from C2 cells. These results provide valid evidence that C2 early T cells were endogenous retrovirus- transformed leukemia cells.
