摘要
目的:建立天然牛黄HPLC-ELSD特征图谱并与酶促牛黄特征图谱进行比较。方法:采用HPLC-ELSD法,色谱条件:Diamonsil C18柱(250 mm×4.6 mm,5μm);以甲醇和0.1%的醋酸水溶液为流动相,流速0.8 mL.min-1,梯度洗脱,漂移管温度110℃,气体流速2.0 L.min-1;分析时间60 min。结果:在选定的色谱条件下建立了天然牛黄色谱特征图谱共有模式,并对酶促牛黄与天然牛黄的特征图谱进行比较。结论:所建立的天然牛黄HPLC-ELSD特征图谱方法精密度高、重现性好,可用于酶促牛黄的鉴别研究。
Objective：To establish an HPLC-ELSD specific chromatogram analysis method of natural Calculus Bovis with a comparative study of Calculus Bovis cultivated by glucuronidase.Methods：An HPLC-ELSD analysis was carried out on a Diamonsil C18 column（250 mm×4.6 mm,5 μm）with the mobile phase consisting of methanol（A）-0.1% acetic acid aqueous solution（B）in gradient mode.The flow rate was 0.8 mL·min-1.The temperature of drift tube was 110 ℃,flow rate of the gas was 2.0 L·min-1and the analytic time was 60 min.Results：Under the above chromatographic conditions,the mutual mode of HPLC specific chromatograms was set up,Calculus Bovis cultivated by glucuronidase had a similar specific chromatogram with the natural Calculus Bovis.Conclusion：The method is in good precision and reproducibility,which can be available for the identification of Calculus Bovis cultivated by glucuronidase.
