RANTES基因启动子区-28C/G多态性与2型糖尿病的相关性研究

摘要 目的探讨正常T细胞表达和分泌的活性调节蛋白(RANTES)基因启动子区-28C/G多态性与T2DM及其慢性并发症相关危险因素的相关性。方法运用PCR-RFLP技术对178例T2DM患者(T2DM组)和67例正常对照者(NC组)的RANTES基因-28C/G多态性进行检测。结果 NC组和T2DM组间基因型和等位基因频率差异无统计学意义(P>0.05);RANTES基因-28G等位基因携带者血浆HDL-C浓度较非G等位基因携带者低且差异有统计学意义(P<0.05);Logistic回归分析:RAN-TES基因启动子区-28C/G多态性与HDL-C水平无关(P=0.228,OR=1.777),BMI与HDL-C异常呈正相关(P=0.000,OR=2.562)。结论 RANTES基因启动子区-28C/G多态性可能不是昆明地区汉族T2DM发生的危险因素,超重和肥胖可能是T2DM患者HDL-C异常的危险因素。 Objective To investigate the associatoin of RANTES (regulated on activation, normal T-cell expressed and secreted) gene promoter -28C/G polymorphism with type 2 diabetes mellitus (T2DM) and risk factors related to chronic complications. Methods RANTES promoter -28C/G polymorphism was detected by PCR-RFLP in 178 T2DM patients and 67 normal controls (NC). Results There were no statistical differences in genotype and allele frequencies between NC group and T2DM group. The level of HDL-C in patients with RANTES- 28G-positive genotype was significantly lower than with -28G-negtive genotype ( P〈0. 05 ). Logistic regression analysis showed RANTES promoter -28C/G polymorphism was not associated with the level of HDL- C. BMI was positivly correlated with HDL-C abnormality. Conclusion RANTES promoter -28C/G polymorphism may not be a risk factor for T2DM, while, overweight and obesity may be risk factors for HDL-C abnormality of patients with T2DM in Kunming Han people.
