摘要
目的:应用建立的整合荧光素酶的稳定细胞克隆构建裸鼠肺癌模型,并用活体成像技术检测肿瘤成瘤及变化。方法:细胞计数分析其生长特性,体外测定不同数量的细胞的发光强度;尾静脉注射接种BLAB/c nu/nu裸鼠,活体成像系统监测裸鼠体内肿瘤的生长及转移,肿瘤组织切片验证移植瘤的病理特性。结果:在稳定表达荧光素酶的SPC-A-1-luc+细胞基因组中能有效扩增到荧光素酶基因;该细胞系具有与母细胞相同的生长曲线,体外检测发现其发光强度与细胞数量呈正相关;尾静脉接种裸鼠后肺部成瘤率为100%,且信号强度随时间递增。结论:建立了稳定整合Luciferase基因的肺腺癌细胞株SPC-A-1-luc+稳定克隆,尾静脉接种后可有效构建肺部肺腺癌成瘤模型,并可动态监测肿瘤的生长及转移。
Objective To establish a mouse lung cancer model in lung with stable cell line integrated with luciferase gene and monitor the tumor by an in vivo imaging system.Method A lung adenocarcinoma model of BALB/c nu/nu nude mouse was established by intravenous injection with the stable cell line.The growth and metastasis of the tumor was monitored by in vivo imaging system.The tumor in lung was verified by tissue sections with Hematoxylin and Eosin(HE) staining.Results The luciferase gene can be amplified in genomic DNA of SPC-A-1-luc+,good correlation between light emission and cell number was observed in SPC-A-1-luc+ cell line,and the light emission was detected at lung in all BALB/c nu/nu nude mouse,but light emission of head and lower abdomen was detected in part of mice.Conclusion A mouse model of lung cancer in lung was successfully established,and the model can be used for in vivo imaging.
