依达拉奉对脑出血大鼠血肿周围组织Caspase-3和Pdcd-5蛋白表达的影响 被引量:11

Effect of Edaravone on Expression of Caspase-3 and Pdcd-5 Protein after Intracerebral Hemorrhage in Rats

摘要 目的:探讨依达拉奉对脑出血大鼠血肿周围组织Caspase-3和Pdcd-5蛋白表达的影响。方法:102只健康雄性Wistar大鼠,随机分为假手术组6只、脑出血组48只和依达拉奉治疗组48只。后2组又分为3 h、6 h、12 h、1 d、2 d、3 d、5 d、7 d 8个亚组,每组6只。采用大鼠尾动脉自体不凝血50μL注入尾状核区建立脑出血模型。免疫组化法检测Caspase-3和Pdcd-5蛋白表达变化。结果:与脑出血组相比,依达拉奉治疗组于出血后3 h血肿周围组织Caspase-3和Pdcd-5蛋白表达减少,但差异无统计学意义(P>0.05),出血后6 h、12 h、1 d、2 d、3 d、5 d、7 d,2组的Caspase-3和Pdcd-5蛋白差异有统计学意义(P<0.05或P<0.01)。结论:依达拉奉可抑制Caspase-3和Pdcd-5蛋白表达,对脑出血后脑组织损伤有保护作用。 Objective: To investigate the effect of edaravone on the expression of Caspase-3 and Pdcd-5 protein following intracerebral hemorrhage (ICH) in rats. Methods: One hundred and two healthy Wistar rats were randomized into sham group (n=6), ICH group (n=48) and edaravone treatment group (n=48). The later two groups were randomized into 8 subgroups respectively: 3 h, 6 h, 12 h, 1 d, 2 d, 3 d, 5 d and 7 d groups after ICH, and 6 rats in each group. ICH models were prepared by injecting 50μL of autogenous blood from the central tail artery into the right caudate nucleus of rats. The changes of Caspase-3 and Pdcd-5 protein were observed by immunohistochemistry respectively. Results:Compared with ICH group, expressions of Caspase-3 and Pdcd-5 protein were decreased 3 h after ICH in edaravone treatment group, but there was no sta- tistical difference between two groups (P 〉 0.05). Expressions of Caspase-3 and Pdcd-5 protein were significantly different between groups including 6 h, 12 h, 1 d, 2 d, 3 d, 5 d and 7 d(P 〈 0.05 or P 〈 0.01). Conclusion: There was an inhibitory effect of edaravone on expressions of Caspase-3 and Pdcd-5 protein following intraeerebral hemorrhage in rats, which protected intracerebral hemorrhage injury.
