The Effect of Lutein Pretreatment on Cisplatin-Induced Hepatic Ultrastructure Damage in Rats

摘要 目的:观察叶黄素(LU)对顺铂(CP)损伤大鼠肝组织超微结构的影响。方法:将24只雌性SD大鼠随机分为生理盐水组(NS组)、顺铂化疗组(CP组)和LU干预组(LU+CP组),每组8只。NS组及CP组大鼠灌胃玉米油2 mL/kg,LU+CP组大鼠灌胃20 g/L的LU油溶液2 mL/kg。灌胃7 d后,CP组和LU+CP组腹腔注射1 g/L的CP 5 mg/kg,NS组腹腔注射等容量生理盐水。注射CP后第5天剖取肝脏,在光镜、透射电镜下观察大鼠肝组织形态结构改变。结果:LU+CP组的CP所致的肝细胞质空泡化及肝门管区纤维化等改变较CP组减轻,同时LU+CP组CP引起的线粒体嵴结构断裂或消失、粗面内质网呈片层状及染色质边集于核膜下等超微结构改变也较CP组减轻。结论:预防性喂饲LU可对CP所致的大鼠肝脏病理及超微结构损伤产生一定的保护作用。 Objective: To observe the effect of lutein (LU) on the hepatic ultrastructure damage induced by cisplatin (CP) in rats. Methods: Twenty-four adult SD rats were randomly divided into normal saline group (NS), cisplatin treatment group (CP) and LU+CP group (n=8 for each group). Rats in control group and CP group were administered corn oil orally at 2 ml/kg. Rats in LU+CP group were administered 20 g/L LU solution at 2 mL/kg for 7 days. Then 1 g/L CP 5 mg/kg were injected intraperitoneally in rats of CP group and LU+CP group, while those in control group were given the same volume of normal saline. Five days after injection all rats were killed to dissect liver. The hepatic morphological structures were observed under light microscope and transmission electron microscope (TEM). Results: LU could alleviate the hepatic histopathological damages induced by CP, such as the cytoplasm vacuolated and liver portal canal fibrosis. As well, hepatic uhrastructural damages were markedly relieved in LU protection group, which included mitochondrial cristae breakdown or loss, the endoplasmic reticulum appeared lamellar, and the nuclear chromatin condensed and marginated. Conclusion: A preventative administration of LU might protec, t hepatic pathological structure and uhrastructure from damages induced by cisplatin in rats.
