葛根素注射液治疗糖尿病性骨质疏松的实验研究 被引量:11

摘要 目的:探讨葛根素注射液对糖尿病骨质疏松的保护作用。方法:30只SD大鼠随机分为正常对照组、糖尿病组和糖尿病葛根素治疗组(n=10)。糖尿病组和糖尿病葛根素治疗组大鼠经腹腔注射链脲佐菌素(65mg/kg)制作糖尿病模型,成模后糖尿病葛根素治疗组每天腹腔注射葛根素注射液80mg/kg,正常对照组和糖尿病组注射等量生理盐水。成模6周后麻处死大鼠,分离各组大鼠的左侧股骨,于小动物放射成像系统上,测其近段骨密度,HE染色进行形态学观察。结果:成模6周后,糖尿病组骨密度显著低于对照组,糖尿病大鼠其股骨头表面成骨细胞与对照组比较显著减少,空骨陷窝明显增多,而葛根素治疗组上述指标均明显改善。结论:高糖可影响骨组织代谢及成骨细胞的增殖分化,造成糖尿病骨质疏松症,而葛根素对骨质疏松的进程可起到一定的保护作用。 Objective:To explore the protective effect of puerarin on diabetic osteoporosis rats.Methods:Thirty male SD rats were divided into three groups,including control group(intraperitoneally injected saline 6 mL/kg),diabetic group(intraperitoneally injected STZ 65 mg/kg)and puerarin treatment group(intraperitoneally injected STZ 65 mg/kg,and intraperitoneally injected puerarin 80 mg/kg/day for 6 weeks).After 6 weeks,the rats were isolated left femur,the bone mineral density of proximal femur was measured by the small animal x-ray imaging device,and the morphological changes were also observed.Results:Bone mineral density in diabetic group was significantly lower than that in control group.Haematoxylin-eosin(HE)staining of femur revealed the femoral head surface of osteoblasts was a significant reduce and empty lacunae increased.But the osteoporosis performance was significantly improved in the puerarin group rats.Conclusion:High blood sugar can affect bone metabolism and osteoblasts proliferation and differentiation,resulting in diabetes,and the puerarin may play a protective role in the process.
