摘要
目的探讨扶正抑瘤方(FYG)与5-FU联用对凋亡相关因子及5-FU靶酶TS基因、蛋白表达的影响。方法建立H22肝癌ICR小鼠皮下移植瘤模型,分模型组、5-FU组、FYG组、5-FU+FYG 4组,连续给药FYG(3.6 g/kg.d)和5-FU(10 mg/kg.d)5 d,免疫组化方法检测肿瘤组织中凋亡相关因子(Bax,Bcl-2)和TS的蛋白表达情况,RT-PCR检测凋亡相关因子(Bax,Bcl-2,p53,C-myc)和TS的基因表达。结果与模型组相比,其它3组小鼠肿瘤组织中Bax蛋白表达显著上调(P<0.05 or P<0.01);FYG组和5-FU+FYG组小鼠肿瘤组织中Bax基因表达明显上调(P<0.05or P<0.01);5-FU组和5-FU+FYG组小鼠肿瘤组织中野生型p53基因表达显著上调(P<0.05 orP<0.01)。结论 FYG与5-FU联合应用可上调凋亡相关因子蛋白及基因的表达,但对TS几乎没有影响。
Objective To explore the effects of Fuzheng Yiliu decoction(FYG) and 5-FU on gene and protein expressions of apoptosis related factors and TS of H22 tumor-bearing ICR mice.Methods The models of H22 tumor-bearing ICR mice were established and mice were divided into model group,5-FU group,FYG group and 5-FU+FYG group.After successive administration for 5 days,the protein expressions of Bax,Bcl-2 and TS were detected by immunohistochemical method,and the gene expressions of Bax,Bcl-2,p53,C-myc and TS were detected by RT-PCR.Results Compared with model group,the protein expression of Bax in tumor tissue of mice in other groups was respectively up-regulated(P0.05 or P0.01),the gene expression of Bax in FYG group and 5-FU+FYG group was respectively up-regulated(P0.05 or P0.01),and the gene expression of wild type p53 in 5-FU group and 5-FU+FYG group was respectively up-regulated(P0.05 or P0.01).Conclusion FYG and 5-FU could up-regulate the gene and protein expressions of apoptosis related factors,but have almost no effect on TS.
