透骨消痛胶囊中两个部位配比对大鼠膝骨性关节炎炎症因子表达的影响

Effect of Compatibility of Two Parts from Tougu Xiaotong Capsule on Inflammatory Factors Expression in Rats with Knee Osteoarthritis

目的探讨透骨消痛胶囊2个有效部位不同配比对骨性关节炎炎症因子表达的影响。方法清洁级SD大鼠90只,采用关节腔注射木瓜蛋白酶复制大鼠膝骨性关节炎模型,随机分为9组,正常组、模型组(生理盐水)、实验1-7组(苷类部位:生物碱部位分别为10∶0,10∶1,10∶3,10∶6,10∶10,1∶10,0∶10),分别干预4周后,采用放射免疫法测定关节液IL-1、IL-6、TNF-α、MMP-3、HA、NO含量。结果透骨消痛胶囊苷类部位与生物碱部位不同配比均能有效降低骨性关节炎关节液中IL-1、IL-6、TNF-α、MMP-3、NO含量,提高骨性关节炎关节液中HA含量。结论透骨消痛胶囊的苷类部位与生物碱部位不同配比均能有效抑制膝骨性关节炎的炎症反应,其中苷类部位与生物碱部位为10∶6的效果较好。

Objective To explore the effect of compatibility of two parts from Tougu Xiaotong capsule（TGXTC） on the inflammatory factors expression in rats with knee osteoarthritis（KOA）.Methods 90 clean SD rats were randomly divided into normal group,model group and experiment group I-Ⅶ.KOA rat models were established by intra-articular injection of papain solution.The normal group and model group were treated with physiological saline,and experiment groups were treated with saponins and alkaloids in different ratios for 4 weeks,then the levels of IL-1,IL-6,TNF-α,MMP-3,HA and NO in synovia fluid were detected by radioimmunity assay.Results The compatibility of saponins and alkaloids in different ratio from TGXTC could down-regulate the levels of IL-1,IL-6,TNF-α,MMP-3 and NO,and up-regulate the level of HA in synovia fluid of KOA patients.Conclusion The compatibility of saponins and alkaloids in different ratio from TGXTC could effectively inhibit the inflammation of KOA.Among the ratios,the ratio of saponins and alkaloids with better effect is ten to six.
