胆管癌神经浸润转移与DPC4神经细胞粘附分子表达偶联的关系 被引量:4

Relationship between perineural invasion and the coupling expression of DPC4 with NCAM in cholangiocarcinoma

摘要 目的 研究DPC4基因和神经细胞粘附分子(neuralcelladhesionmolecule,NCAM)分子表达的偶联性与胆管癌神经浸润转移的关系。方法 以RT PCR技术扩增 18例新鲜胆管癌标本RNA ,32 P标记DPC4 ,NCAM作探针 ,作斑点杂交 ,放射自显影图像分析确定 3种分子表达强度 ,建立DPC4和NCAM表达关系的数学模型。结果 DPC4基因表达强度随胆管癌恶性程度的增加而降低 ,NCAM则升高。数学关系模型为Y =0 .5 95 5 / (- 0 .0 3 3 9+X) ,显示两者呈负相关(P <0 .0 5 )。结论 DPC4基因可能与NCAM基因在功能上有负偶联性 ,DPC4可能通过某种信号转导途径调控NCAM基因的表达 ,从而影响胆管癌向神经浸润转移。 Objective\ To study the relationship between perineural invasion and the coupling expression of DPC4 with neural cell adhesion molecule (NCAM) associated with nerve damage. Methods\ Eighteen frozen cholangiocarcinoma specimens were taken directly from the patients. Total RNA was extracted from the specimens for RT PCR. Dot blotting for the expression of DPC4, NCAM and β actin was used by probes of those three kinds of molecules labeling with 32 P. The quantity expression of the molecules was measured by image analysis technique. Results\ The expression of NCAM was increased, while that of DPC4 was decreased with the malignancy of the cancer. It was found by regression analysis that they bore a remarkable negative correlation. Conclusion\ DPC4 gene might modulate the expression of NCAM associated with perineural invasion and metastasis in cholangiocarcinoma through a signal transduction pathway. It is suggested that the negative coupling expression of DPC4 and NCAM might be an unique character and might play a very important role in the perineural invasion of cholangiocarcinoma.
