摘要
目的探讨组蛋白乙酰化酶抑制剂丙戊酸钠(VPA)对人宫颈癌Hela细胞增殖和凋亡的作用及其可能机制。方法不同浓度的VPA处理Hela细胞,应用Western blot法检测细胞内组蛋白H4乙酰化水平,荧光定量反转录聚合酶链式反应(RT-PCR)方法检测p21基因mRNA表达,四甲基偶氮唑蓝法(MTT)检测细胞增殖,流式细胞术检测细胞凋亡率。结果经VPA作用后,人宫颈癌Hela细胞内组蛋白H4乙酰化水平增加,p21基因mRNA表达增加;同时细胞生长受到抑制,细胞凋亡增加。结论 VPA通过提高组蛋白乙酰化水平,增加p21基因表达,抑制人宫颈癌Hela细胞的增殖和诱导细胞凋亡。
Objective To investigate the effects of histone acetylation enzyme inhibitor of valproic acid sodium(VPA) on proliferation and apoptosis of human cervical carcinoma Hela cells and the possible mechanism. Methods Hela cells were treated with different concentrations of VPA.The acetyl level of histone H4 was detected by Western blot,the mRNA expression of p21 gene was detected by real time reverse transcription polymerase chain reaction(RT-PCR).The cellular growth and cell apoptosis rate were estimated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) and flow cytometry respectively. Results After treatment with VPA,the acetyl level of histone H4 and mRNA expression of p21 gene increased in Hela cells respectively.Meanwhile,the cell growth activity decreased and cellular apoptosis increased. Conclusion The VPA could effectively inhibit the cellular proliferation and induce apoptosis of cervical carcinoma Hela cells by increasing the acetyl level of histone and enhancing p21 gene expression.
