摘要
采用平板对峙培养法和PAGE技术研究了12个金针菇菌株间的亲缘关系。平板对峙培养法研究结果表明:12个菌株间存在不同程度的拮抗现象,无拮抗或拮抗效果不明显的菌株组合为:1号和6号、2号和4号、7号和10号、3号和5号。基于12个菌株的酯酶同工酶图谱的聚类分析结果表明,12个菌株在遗传距离为D=0.39处明显地聚成5组。其中,Ⅰ组为1号和6号,呈现Ec、Ed、Ei、Ek、El特征带;Ⅱ组为2号和4号,呈现Ec、Ed、Ef~Ei、El特征带;Ⅲ组为7号、8号、9号和10号,呈现Ed、Eh、Ei和El特征带;Ⅳ组为3号、5号和11号,呈现Ea、Ed~Ei、El特征带;Ⅴ组为12号,呈现Ed、Ee、Ei和El特征带。拮抗关系法和酯酶同工酶图谱聚类分析结果基本一致。此外,12个金针菇菌株的酯酶同工酶图谱中,谱带d、i、l为12个菌株所共有,说明这3条谱带为金针菇的标志性谱带,其相对迁移率分别为0.479、0.744、0.905。
The phylogenetic relationships among the 12 types of Flammulina velutipes strains were identified by the analysis of antagonistic action and PAGE.There were different degrees of antagonism between these strains.The test results of antagonistic action suggested that No.1 and No.6,No.2 and No.4,No.7 and No.10,No.3 and No.5 were of the closest relationship,respectively.There were no antagonism or no obvious antagonism each other.The dendrogram of esterase isozyme of the 12 types of Flammulina velutipes strains showed that the strains were clustered into five groups using similarity coefficient of 0.39 as a cut-off point.Group one consisted of No.1 and No.6 strain with the characteristic bands of Ec,Ed,Ei,Ek,El;group two consisted of No.2 and No.4 strains with the characteristic bands of Ec,Ed,Ef~Ei,El;group three consisted of No.7,8,9,10 strains with the characteristic bands of Ed,Eh,Ei,El;group four consisted of No.3,5,11 strains with the characteristic bands of Ea,Ed~Ei,El.The five group No.12 with the characteristic bands of Ed、Ee,Ei,El.The test results of antagonistic action and the analysis of the dendrogram of esterase isozyme were basically the same results.In addition,the iconic bands of Ed,Ei,El with different migrating rate(0.479,0.744,0.905) were found in all the needle mushroom strains.
