蒙药珍宝丸药物血清对雪旺细胞增殖的影响

Effect of the rat's serum with Zhenbao Pill on Schwann cells proliferation

目的观察蒙药珍宝丸药物血清对体外培养雪旺细胞增殖的影响。方法选用新生24 h SD大鼠,取双侧坐骨神经,分离、培养并纯化雪旺细胞,用S-100免疫细胞化学染色法鉴定雪旺细胞,将纯化的雪旺细胞分别用胎牛血清、不同给药剂量大鼠血清、以及不含药大鼠血清培养,并分别于培养的第1天、第4天以及第7天用MTT法观察各组雪旺细胞增殖情况。结果 (1)S-100免疫染色结果显示纯化后的雪旺细胞纯度为93.8%。(2)MTT法测得培养第7天时高剂量含药血清组OD值明显高于正常剂量含药血清组和不含药大鼠血清组OD值(P<0.05),而全过程中胎牛血清组OD值明显高于不含药大鼠血清组OD值(P<0.05)。结论蒙药珍宝丸药物血清在适度条件下可促进雪旺细胞的增殖。

AIM To investigate the effect of the rat＇s serum with Zhenbao Pill on Schwann cells in culture. METHODS The sciatic nerve from SD rats （24 h-born） was cultured in vitro and purified. S-100 immunhistochemical staining was performed to identify the Schwann cells. The purified Schwann cells were treated with fetal calf serum, and the different dose rat＇s serum with and without Zhenbao Pill. MTT was used to observe the viability of the Schwann cells on the first, fourth and seventh day. RESULTS Based on S-100 immunhistochemical staining, greater than 93.8% of Schwann cells in culture was identified with certainty. MTT coloritric assay revealed that OD value in high dose rat＇s serum with Zhenbao Pill elevated compared with low dose rat＇s serum with Zhenbao Pill and without Zhenhao Pill on the seventh day （P 〈 0.05 ）. OD value treated with fetal calf serum was obviously greater than OD value without rat＇s serum. CONCLUSION The rat＇s serum with Zhenbao Pill at the optimal concentration is effective in increasing the proliferation of Schwann cells.
