摘要
目的研究搭载兔骨髓基质干细胞(BMSCs)的温敏型壳聚糖/β-甘油磷酸钠(C/GP)水凝胶复合支架对兔桡骨骨折的治疗疗效。方法将体外培养、扩增的兔BMSCs与C/GP水凝胶混合植入兔桡骨骨折间隙内作为实验组与空白对照组进行比较。通过x线片、生物力学检测及组织形态学检测等方法对骨折愈合情况进行评定。结果术后第4周和第12周实验组通过X线片反应出的骨愈合结果明显强于对照组;术后第12周生物力学检测实验组中最大负荷、抗弯曲强度及载荷/位移等参数与对照组比较差异均有统计学意义(P〈0.05),而弹性模量两组比较差异无统计学意义(P〉0.05);术后第12周,通过组织形态学观察实验组成骨及塑形等方面明显优于对照组。结论搭载BMSCs的温敏型C/GP水凝胶复合支架材料可促进骨折愈合。
Objective To investigate the effect of thermosensitive chitosan /β-glycerol phosphate (C/GP) hydrogel composite scaffolds embarking rabbit bone marrow stromal cells (BMSCs) on the healing of rabbit radial fractures. Methods The mixture of C/GP hydrogel and rabbit BMSCs cultured and proliferated in vitro was transplanted into the fracture interspace in an experimental group in comparison with a blank control group. X-ray, biomeehanical measurement and histochemical assay were conducted to assess the effect of scaffolds on fracture healing between the 2 groups. Results At 4 and 12 weeks post-treatment, X-ray showed the fracture healing in the experimental group was significantly better than in the control group. At 12 weeks postreatment, biomechanical measurement showed significant differences regarding maximum load, flexural strength and load/displacement parameters between the experimental and the control groups ( P 〈 0. 05), but no significant difference in elastic modulus parameters ( P 〉 0.05). At 12 weeks post-treatment, histochemical assay showed the osteogenesis and bone moulding capability were obviously better in the experimental group than in the control group. Conclusion The thermosensitive C/GP hydrogel composite scaffolds embarking BMSCs can promote fracture healing.
