淋浴热水军团菌传统培养与实时荧光PCR技术的应用 被引量:1

Application of Conventional Culture and Real Time PCR Method in Detection of Legionella in Shower Warm Water

摘要 目的利用传统的检验方法与实时荧光PCR技术相结合检测军团菌。方法按照卫生部《公共场所集中空调通风系统卫生规范附录A》(2006)的要求,于2010年10月—2011年12月,对深圳市6家宾馆酒店86件淋浴热水进行军团菌检测。采用传统细菌分离培养法进行分离鉴定,采用实时荧光PCR法加以验证。结果在86件淋浴热水中,20件检出嗜肺军团菌,嗜肺军团菌阳性率为23.3%,其中,Lp1、Lp3、Lp6血清型分别占65%,10%,25%;实时荧光PCR法鉴定出1件淋浴热水为非嗜肺的军团菌属。结论深圳市宾馆酒店淋浴热水存在军团菌的污染;传统的检验方法与荧光PCR技术的检测结果具有较高的一致性。 Objective To detect Legionella in shower warm water by conventional culture method and real-time fluorescent PCR. Methods According to the Sanitation Criteria for Central Conditioners in Public Buildings, a total of 86 samples of shower warm water were detected,isolated and cultured with conventional method in October 2010-December 2011. Real time fluorescent PCR method was used to confirm the results. Results A total of 20 strains of Legionella pneumophila and a strain of non-Legionella pneumophila were found in the 86 samples.The positive rate of LegioneUa pneumophila was 23.3%,Lpl, Lp3, Lp6 were 65%, 10% ,25%. Conclusion Legionella exists in shower warm water at hotels in Shenzhen. The conventional culture and real-time fluorescent PCR has high consistency.
