摘要
目的对链霉菌ZZ027和ZZ021所产抗耐甲氧西林金黄色葡萄球菌(MRSA)次级代谢产物的结构类型进行快速鉴定。方法用2A培养基对两链霉菌菌株进行发酵;再采用HPLC微分离技术和琼脂扩散法对抗MRSA色谱峰进行定位,然后采用TLC对发酵液中抗MRSA次级代谢产物进行分离与制备,所得抗MRSA组分经薄层显色、IR和13C-NMR谱测试分析,鉴定其结构类型。结果链霉菌ZZ027和ZZ021抗MRSA组分的薄层显色、IR和13C-NMR谱特征均与大环内酯类化合物的相关表征一致。结论链霉菌ZZ027和ZZ021所产抗MRSA次级代谢产物为大环内酯类化合物。
OBJECTIVE To quickly identify the structure types of anti-methicillin-resistant Staphylococcus aureus(anti-MRSA) secondary metabolites produced by Streptomyces sp.ZZ027 and Streptomyces sp.ZZ021.METHODS Two strains were fermented with 2A medium,and anti-MRSA chromatographic peaks were localized by HPLC micro-separation technology and agar diffusion method;anti-MRSA ingredients were isolated and prepared by TLC method,and their structure types were deduced by TLC chromogenic reaction,IR and 13C-NMR spectra.RESULTS The characters of chromogenic reaction,IR and 13C-NMR spectra of anti-MRSA ingredients produced by Streptomyces sp.ZZ027 and Streptomyces sp.ZZ021 were consistent with those of macrolides.CONCLUSION Anti-MRSA secondary metabolites produced by Streptomyces sp.ZZ027 and Streptomyces sp.ZZ021 were macrolides.
