摘要
目的 研究DT390基因与VEGF人外显子7基因晤后对胃癌细胞的杀伤作用,探索毒素晤基因治疗胃癌的有效途径,方法 构建携带DT90-VEGF165或DT390-VEGFexon7融合基因的真核表达载体,用脂质体介导转染胃癌细胞SGC7901,观察胃癌细胞的形态变化,结果 5μg趑 核表达构的转染5×10^5个胃癌细胞/孔(24孔板)96h后发现,转素融合基因的细胞90%死亡,而对照细胞形态正常。
AIM To study the killing effect of the fusion geneconsisting of DT390-VEGF165 or DT390-VEGFexon7 on humangastric carcinoma cell.METHODS The eukaryotic expression vector carryingDT390-VEGF165 or DT390-VEGFexon7 gene was constructed.The eukaryotic expression constructs were introducedinto gastric carcinoma cell SGC7901 by means oflipofectamine-mediated gene transfer procedure. Thecytolytic effect of the chimeric gene was registered byphotomicrography.RESULTS The most gastric carcinoma cells of 5×105cells/ well were cytosis after transferred with 5 μgeukaryotic expression constructs 96 hours later, but thecontrol cells were normal.CONCLUSION The chimeric gene can be expressed in thegastric carcinoma cell and result in obvious antitumoreffects.
