摘要
目的建立高效液相色谱测定人体血浆中米卡芬净钠(抗真菌药)浓度的方法。方法血浆样品经甲醇沉淀蛋白提取分离,色谱柱为CAPCELL PAK C1柱,流动相为甲醇-10 mmol.L-1醋酸铵(57∶43,v/v),流速为0.2 mL.min-1。结果米卡芬净钠在0.2~25.0μg.mL-1线性良好,最低定量限为0.2μg.mL-1,日内RSD均≤4.1%,日间RSD均≤5.0%,提取回收率RSD均≤6.8%。结论本方法灵敏、快速、准确,可用于米卡芬净钠药代动力学测定。
Objective To establish a HPLC method for determining micafungin concentration in human plasma.MethodsThe plasma samples were extracted by methanol.The separation was performed on CAPCELL PAK C1 column with mobile phase consisted of methanol-10 mmol·L-1 CH3COONH4(57∶43) at the flow rate of 0.2 mL·min-1.ResultsDetermination of micafungin has good linearity in the 0.2-25.0 μg·mL-1,lower limit of quantification is 0.2 μg·mL-1.The intra-day deviation less than 4.1%,inter-day deviation less than 5.0%.ConclusionThe established HPLC method sensitive,fast,accurate and suitable for determination of micafungin in human plasma.
