水杨酸钠降低庆大霉素所致耳蜗自由基含量的实验研究 被引量:7

An Experimental Study on Salicylate Preventing Exessive Free Radicals in Cochlea From Generated by Gentamicin Injection

摘要 目的 采用电子顺磁共振法 (electronparamagneticresonancespectroscopy ,EPR)观察水杨酸钠对庆大霉素注射过程中耳蜗组织羟自由基含量增加的预防作用。方法 耳廓反射良好的豚鼠随机分成 3组 ,庆大霉素组、庆大霉素 +水杨酸钠组和正常对照组。取标本前和标本制备时向耳蜗组织加入自由基捕捉剂 5 ,5 -dimethyl -pyrroline - 1-oxide(DMPO) ,用EPR直接检测耳蜗组织中自由基含量。结果 正常对照组耳蜗组织可测出较弱的羟(OH)自由基信号 ,庆大霉素注射后 ,自由基信号明显增强 (P <0 .0 1) ,水杨酸钠和庆大霉素同时注射 ,自由基信号明显减弱 (P <0 .0 1)。结论 加入自由基捕捉剂DMPO ,用EPR直接检测耳蜗组织中自由基含量是一种较有效的方法 ; Objective To investigate the effed of salicylate in preventtng excessive free radicals in cochlea from generated by Gentamicin injection.Methods The animals were arranged in three groups(Gentamicin;salicylate and Gentamicin;control).5,5-dimethyl-pyrroline-l-oxide(DMPO),a spin trap agent,was injected into the cochleas and added in cochlea homogenates.Electron paramagnetic resonance spectroscopy(EPR)was used to provide a direct measure of oxygen radical production-hydroxyl radical in guinea pig's cochlea.Results DMPO-OH adduct coulel be detected in normal cochlea by EPR.There were only weak signals in control group,after animals were injected by Gentamicin for three days,the EPR signals became stronger(P<0.01)when animals were injected with Gentamicin and salicylate simultaneously,signals from free radicals in cochlea weakened obviously(P<0.01).Conclusion EPR,combined with spin trapping,is an effective method to detect free radicals in cochlea,and salicylate can partly prevent excessive free radicals from generatied by Gentamicin injection.
