摘要
探讨卵巢癌细胞凋亡过程相关基因及其蛋白表达的变化 ,为解决卵巢癌化疗耐药提供理论依据。方法 :应用不同浓度顺铂处理HO - 8910细胞 ,HE染色观察细胞形态改变 ,琼脂糖凝胶电泳观察DNA降解 ,SP免疫组化染色观察细胞凋亡过程中P53、Bcl - 2蛋白表达的变化。结果 :由于使用顺铂 ,HO - 8910细胞呈凋亡改变 ,DNA琼脂糖凝胶电泳呈典型的凋亡特征。在细胞凋亡的同时 ,P53蛋白表达逐渐升高 ,Bcl- 2蛋白表达逐渐降低 ,二者的变化分别和凋亡细胞数呈正、负相关性。结论 :顺铂通过细胞内P53、Bcl - 2蛋白表达的变化 ,诱导人卵巢癌细胞凋亡是其抗肿瘤的机制之一。卵巢癌细胞对顺铂耐药的产生可能和P53、Bcl- 2蛋白表达有关。
Objective:To elucidate the mechanism of treatment on ovarian carcinoma by cisplatin and explore the change of genes and their expression proteins in the apoptosis process induced by cisplatin Methods:HO-8910 cells were treated with vary concentration of cisplatin Morphological changes(HE stain) were observed under light microscope DNA degradation was assayed by electrophoresis on ayarose gel The change of expression of P53、Bcl-2 proteins was examined immunohistochemically(SP) Results:Under the influence of cisplatin,HO-8910 cells exhibited changes of apoptosis in morphology The expression of P53 protein increased and that of Bcl-2 protein decreased There were positive correlation and negative correlation between apoptosis and the expression of P53、Bcl-2 respectively Conclusions:Inducing apoptosis is one of the primary mechanism of anti-cancer chemotherapy of cisplatin The expression of P53、Bcl-2 protein is involved in the mechanism of apoptosis and resistance of ovarian carcinoma to chemotherapeatic-drugs
