胃癌中Cyclin D1和p53蛋白的表达及意义

Expression and significance of Cyclin D1 and p53 in gastric cancer

目的 检测 72例胃癌和癌旁组织及 2 0例正常胃粘膜中的CyclinD1和p5 3蛋白 ,探讨它们在胃癌发生、发展中的作用及意义。方法 采用链菌素亲生物素———过氧化物酶法 (SP法 )。结果 胃癌组织中CyclinD1和p5 3蛋白总阳性表达率分别为 47.2 2 %和 5 2 .78% ,与癌旁组织或正常对照胃粘膜组织比较均有非常显著性差异 (P <0 .0 1) ;二者均与胃癌分化程度有关 ,其中p5 3蛋白表达与肿瘤深度 ,淋巴结转移 ,远处转移等因素有明显相关 (P <0 .0 1) ,但CyclinD1表达与之无关 (P >0 .0 5 )。胃癌中CyclinD1与p5 3蛋白表达呈正相关 (r =+0 .45 )。结论 CyclinD1与p5 3蛋白过表达与胃癌的发生 ,分化有关 ,其中CyclinD1过表达是胃癌发生过程中一个较早的分子事件 ,而p5 3蛋白过表达是相对较晚的分子事件 ,可做为早期诊断 ,判断预后和胃癌分化程度的一个参考指标。

Objective To detect Cyclin D1 and p53 in gastric mucosa of gastric cancer,and its adjacent tissue and normal gastric tissue and to probe into their roles in the occurrence and development of gastric cancer.Methods SP assay was performed.Results The total positive rates of Cyclin D1 and p53 in gastric cancer were 47.22% and 52.78% respectively,which had significant difference compared with the adjacent tissue to cancer and normal mucosa (P<0.01).Both Cyclin D1 and p53 had relation to the differentiated degree of gastric cancer.P53 related with the depth of tumor,lymphatic metastasis and distal metastasis (P<0.01),while Cyclin D1 didn′t (P>0.05).The expression of Cyclin D1 and p53 had positive relation (r=+0.45)Conclusions The over expression of Cyclin D1 and p53 related with the ocurrence and differentiation of gastric cancer in gastric cancer.The expression of Cyclin D1 was an earlier event.whereas the expression of P53 was a relatively later event in the occurrence of gastric cancer.Both of them could be taken as a reference in the early diagnosis,prediction of prognosis and judgment of differentiated degree of gastric cancer. 〔
