甘氨酸及二甲基亚砜对冷冻保存兔精液活率的影响 被引量:5

摘要 在筛选出稀释液配方和平衡时间的基础上,探讨了不同浓度的甘氨酸及DMSO对冻融后兔精子活率的影响。试验结果表明:(1)在同一浓度的甘氨酸(分别为7.0mg.mL-1、7.5mg.mL-1和8.0mg.mL-1)条件下,解冻后精子活率与DMSO浓度呈负相关,各组彼此差异显著(p<0.05)。(2)当DMSO浓度为7.5℅时,不同甘氨酸浓度对精液冷冻效果差异不显著(p>0.05);当DMSO浓度为10℅时,甘氨酸浓度为7.5mg.mL-1组的解冻后精子活率49.82%最高,其次是7.0mg.mL-1组43.41%,8.0mg.mL-1组37.29%最低,且各组间差异显著(p<0.05);当DMSO浓度为12.5℅时,甘氨酸浓度7.0mg.mL-1组的解冻后精子活率26.69%显著低于7.5 mg.mL-1组和8.0mg.mL-1组(p<0.05),而7.5mg.mL-1组(32.97%)和8.0mg.mL-1组(34.64%)间无显著差异(p>0.05)。综上所述,獭兔精液冷冻保护液中添加7.5mg.mL-1甘氨酸和7.5%DMSO,可得到较好冷冻保存效果。 This study was conducted to investigate the different concentrations of glycine and DMSO on rabbit sperm motility after cryopreservation on the basis of selected diluent formulation and the equilibrium time. The results showed that: (1)In the same concentration of glyeine (7.0,7.5 or 8.0mg·mL^-1) conditions, thawing sperm motility was negatively correlated with the concentration of DMSO in each group, and the differences were significant (p〈0.05).(2)The effects of glycine concentration on the frozen semen was no significant difference with 7.5% DMSO (p〈0.05), also, with 10% DMSO, the thawed sperm live rate of 7.5 mg·mL^-1 glyeine group was 49.82%, whereas 7.0mg·mL^-1glycine group was 43.41%, and 8.0mg·mL^-1 glyeine group was 37.29%. The differences between 7.0mg·mL^-1 and 8.0mg·mL^-1groups was significant (p〈0.05). With 12.5% DMSO, the thawed sperm motility rate of 7.0mg·mL^-1glyeine group was 26.69%, which was significantly lower than 7.5mg·mL^-1and 8.0mg·mL^-1 glyeine group (/9〈0.05), and the difference between 7.5mg·mL^-1(32.97%) glyeine group and 8.0mg·mL^-1 (34.64%) glyeine group was no significant (p 〉0.05). In summary, adding 7.5mg·mL^-1glyeine and 7.5% DMSO can obtain the better Rex Rabbit semen cryopreservation.
