摘要
目的研究人参皂苷Rh1诱导宫颈癌Hela细胞系和白血病K562细胞系的细胞凋亡作用及其机制。方法体外培养Hela细胞和K562细胞,并用Rh1的浓度梯度和时间梯度处理细胞;流式细胞术检测细胞凋亡率;荧光定量PCR检测Bcl-2和Bax基因mRNA表达区别。结果 Rh1以浓度依赖方式诱导Hela细胞核K562细胞凋亡。Bcl-2基因mRNA的表达均出现明显下降,而bax基因,mRNA表达水平均明显上升。结论 Rh1诱导Hela细胞和K562细胞凋亡。Bcl-2表达下降和Bax表达上升是其诱导细胞凋亡的重要机制之一。
Objective To investigate the apoptosis function and mechanism of Ginsenoside Rhl on Hela and K562 cells. Methods Hela and K562 cells were cultured in vitw. Different concentration and time were used to treat the cells and flow cytometry to detect apoptosis rates. Quantitative PCR was applied to detect mRNA expression to identify Bcl-2 and Bax genes. Results The apoptosis induced by Rhl on Hela and K562 were in concentration-dependent manner. Bcl-2 mRNA expression were down-regulated while Bax mRNA expression were up-regulated. Condusion Rhl induced apoptosis of Hela and K562 cells. The down-regulated Bcl-2 and up-regulated Bax are the important mechanism of apoptosis.
