摘要
目的通过不同浓度的IL-1β对小鼠胚胎体外发育的干预,探讨IL-1β在胚胎着床中的作用。方法将IL-1β稀释为1、10、100μg/L等浓度,研究其对2-细胞期小鼠胚胎囊胚形成率及囊胚孵出率的影响,并应用免疫组化方法检测不同浓度IL-1β对着床窗口期小鼠子宫内膜基质金属蛋白酶-9(MMP-9)、细胞黏附分子-1(ICAM-1)蛋白表达的影响。结果经不同浓度的IL-1β干预后,各实验组培养72h的囊胚形成率明显高于对照组,差异均有显著性(χ2=21.050~25.441,P<0.01),各实验组间比较差异无显著性(P>0.05);IL-1β浓度为1、10μg/L组培养96h的囊胚孵出率明显高于对照组,差异均有显著性(χ2=6.705、7.192,P<0.01)。与对照组比较,各实验组MMP-9、ICAM-1蛋白表达量明显增加,差异有显著性(F=149.854、318.097,P<0.01)。结论合适的IL-1β浓度有助于体外培养小鼠胚胎着床。
Objective To explore the intervention of different concentrations of interleukin-1 beta(IL-1β) on in vitro embryonic development and its effect on embryonic nidation in rats. Methods IL-1β was diluted to three concentrations,i.e.1,10,and 100 μg/L,and their effects on 2-cell mouse were observed in terms of the blastocyst formation rate and blastula hatching rate,and their influence on the expressions of MMP-9 and ICAM-1 in endometrium during window phase of nidation were measured immunohistochemically. Results After treatment with different concentrations of IL-1β for 72 h,the blastocyst formation rate was obviously higher than that in the control group,the difference between them being significant(χ2=21.050-25.441,P〈0.01),but the differences among the experimental groups were not significant(P〉0.05).In IL-1β concentration of 1 and 10 μg/L groups,the blastocyst formation rate,after culture for 96 h,was significantly higher than that in the control(χ2=6.705,7.192;P〈0.01).As compared with the control group,the expressions of MMP-9 and ICAM-1 in each experimental group were increased,with a significant difference among them(F=149.854,318.097;P〈0.01). Conclusion A suitable concentration of IL-1β is conducive to embryonic nidation cultured in vitro in rats.
