摘要
目的检测B细胞特异性莫洛鼠白血病病毒插入位点-1(Bmi-1)在非小细胞肺癌(NSCLC)组织的表达,探讨其与上皮间质转化(EMT)的关系。方法采用免疫组化PV-9000两步法检测50例NSCLC组织及30例癌旁相对正常组织Bmi-1、E-钙黏素(E-cadherin)和波形蛋白(Vimentin)的表达情况。结果 Bmi-1、E-cadherin和Vimentin蛋白在NSCLC和癌旁相对正常组织间的表达强度差异均有显著性(Z=-4.413~-2.965,P<0.05)。肺癌组织Bmi-1蛋白与E-cadherin的表达呈负相关(r=-0.416,P<0.05),与Vimentin的表达呈正相关(r=0.356,P<0.05)。结论在NSCLC中,Bmi-1蛋白的过表达伴随着E-cadherin的表达下调和Vimentin的表达上调。Bmi-1可能与NSCLC发生EMT有关,并在其侵袭转移的过程中发挥重要作用。
Objective To detect the expression of B-cell specific Moloney murine leukemia virus insertion site 1(Bmi-1) and investigate its association with epithelial-mesenchymal transition(EMT) in non-small-cell lung cancer(NSCLC). Methods Using PV-9000 immunohistochemical method,the expressions of Bmi-1,E-cadherin and Vimentin in 50 NSCLC samples and 30 correspondingly normal paraneoplastic tissues. Results There were significant differences in the expressions of Bmi-1,E-cadherin and Vimentin between NSCLC and the paraneoplastic normal tissues(Z=-4.413——2.965,P〈0.05).In the cancer tissue,the expression of Bmi-1 was negatively correlated with E-cadherin(r=-0.416,P〈0.05),and positively with Vimentin(r=0.356,P〈0.05). Conclusion In NSCLC,the over expression of Bmi-1 is accompanied by down-regulation expression of E-cadherin and up-regulation expression of Vimentin.Bmi-1 is probably related to EMT in NSCLC and plays a crucial role in the process of invasion and metastasis of this condition.
