玻璃化冷冻与慢速程序冷冻法保存人MⅠ期卵母细胞的效果比较 被引量:2

摘要 目的:探讨玻璃化冷冻与慢速程序冷冻法对人未成熟卵母细胞的存活及胚胎发育潜能的影响。方法:体外受精-胚胎移植中获得的251个MⅠ期卵母细胞随机分3组,第1组94个先玻璃化冷冻保存,第2组63个先慢速程序冷冻保存,两组冻融后的未成熟卵母细胞再进行IVM,用ICSI技术对成熟卵母细胞完成授精,并进行体外胚胎培养。第3组94个MⅠ期卵母细胞先进行IVM,再进行玻璃化冷冻保存,与第1组先玻璃化冷冻保存再IVM的顺序不同,冻融后同样用ICSI技术对获得的成熟卵母细胞完成授精,并进行体外胚胎培养。结果:玻璃化冻融组复苏存活率、体外成熟率、受精率、及卵裂率均高于慢速程序冻融组(P均<0.05);先玻璃化冷冻再IVM与先IVM再玻璃化冷冻的复苏存活率、体外成熟率、可用卵比率、受精率及卵裂率无统计学意义(P均>0.05)。结论:玻璃化冻融人未成熟卵母细胞能获得更好的冷冻保存效果;玻璃化冷冻与IVM的顺序不影响未成熟卵母细胞的利用。 Objective:Comparison of Vitrification and Slow controlled-rate freezing of human immature oocytes.Methods:A total of 251 immature oocytes of MⅠ stage were randomly divided into three groups and froze thawed in different conditions.94 MⅠ stage oocytes were vitrificated,63 MⅠ stage oocytes were Slow controlled rate freezing,After thawing,the immature oocytes were subjected to IVM.ICSI was applied for insemination of the obtained mature oocytes.The rates of survival,maturation,fertilization and cleavage were compared.94 MⅠ stage oocytes IVM before viterification,The rates of survival,maturation,fertilization,and cleavage both from the procedure of vitrification before IVM or IVM before viterfication were compared.Results:The rates of survival,maturation,fertilization and cleavage obviously lowered in Slow controlled rate freezing group(P0.05).No differences were found in both procedure of cryopreservation before IVM or IVM before cryopreservation(P0.05).Conclusion:It was better to Vitrification the MⅠ oocytes.The availability is not different both in the procedure of cryopreservation before IVM or IVM before cryopreservation.
