摘要
斑点热群立克次体的抗原成份极为复杂,它们拥有许多相同的抗原决定簇,也具有各自种特异性的抗原决定簇。本试验使用康氏立克次体(Rickettsia conorii)免疫BALB/C小鼠,取小鼠脾细胞与骨髓瘤细胞系SP2/O-Ag14融合,经克隆筛选,获得2株持续、稳定分泌抗康氏立克次体的单克隆抗体,经微量免疫荧光试验证实为种特异性。应用这2株单克隆抗体结合免疫印迹试验,对康氏立克次体的抗原成份进行初步分析,结果表明康氏立克次体脂多糖样(LPS-like)抗原具有种特异性的决定簇。
Spotted fever group rickettsiae contain many commen cross-reactive antigenic epitopes and specific epitopes. We produced and characterized 2 species of specific murine monoclonal antibodies to Rickettsia conorii, which reacted only with the antigens of Rickettsia conorii by micro-immunofluorescence assay. Western blot showed that these species specific monoclonal antibodies reacted with LPS-like antigen of Rickettsia conorii. Therefore LPS-like antigens of R. Conorii contained specific epitopes.
