ELISA双抗体夹心一步法快速检测粪样中的轮状病毒 被引量:2

摘要 同时将酶标记 McAb与粪样加于抗体包被孔内孵育作 ELISA双抗体夹心一步法,以检测粪样中轮状病毒(RV),发现包被抗体采用兔抗 RV IgG 较抗 RV McAb更为敏感,粪样与酶标记 McAb混合物最适孵育时间为90分钟。经实际粪样测定显示,一步法测得A_(492)值与常规法(二步法)呈直线正相关,两法阳性检出率无显著差异,说明一步法完全可以取代二步法而应用于临床。 Rotavirus (RV) in stools was detected by one -step method with double-antibody sandwish ELISA. The samples and enzymelabeled McAb were incubated simultaneously. It was found that rabbit anti-RV IgG was more sensitive than anti-RV McAb as a capture antibody, and also found that the proper time for incubating samples and enzyme-labeled McAb was 90, minutes. The results showed that there was positively linear correlation in A492 values between one- step and two- step methods, and that no obvious difference in the positive rates of them. Thus, it can be conclused that the one-step method can replace the two-step in clinic.
