摘要
对斯氏狸殖吸虫成虫尿素溶解抗原(P_sAWU)和盐水浸出抗原(P_sAWS)部分特性分析表明,两者氨基酸种类一致,但P_sAWU 酪氨酸和蛋氨酸含量明显高于 P_sAWS;IEP、SDS-PAGA 及 EITB 显示前者活性部分主要在67KD_a 以上,后者则主要在 43KD_a 以下。用于ELISA检测40份该虫病人和感染者血清抗体均阳性,但OD均值及终点滴度 OD 对数均值,P_sAWU 显著高于P_sAWS(P<0.01);检测54份正常人、20份血吸虫病及20份肝吸虫病人血清,均未出现假阳性和交叉反应。此外,两种抗原用于CIEP、IT 检测,效果相同。提示P_sAWU 具有实用价值。
In this paper characteristics and effectiveness of applications of urea soluble antigen of P. skrjabini adult worm (PsAWU) were analysed and compared with that of saline extract antigen of P. skrjabini adult worm (PsAWS). The results of amino acids of determination showed that the kinds of amino acids of two antigens were identical, but the contents of methionine and tyrosine of PsAWU were higher than that of PsAWS. With IEP,SDS-PAGE and EITB it demonstrated that PsAWU and PsAWS contained different antigenic components, with PsAWU mostly greater than 67KD and PsAWS mostly less than 43KD. All of 40 sera from pagumogonimiasis patients and infected individuals detected by ELISA in two antigens were positive,but there were significant differences between OD values and logarithm values of end-points of liters in two antigens(P<0. 01). The results of 54 sera from blood donors,20 sera from schistosomiasis patients and 20 sera from clonorchiasis patients detected by ELISA in the two antigens showed no false and cross-reactions. Moreover, it showed the same effectiveness of applications in PsAWU as did in PsAWS by CIEP and IT. It is suggested that applications of PsAWU is of great value.
