摘要
目的探讨雷公藤甲素对CD4+、CD8+T细胞凋亡的作用。方法分离后的正常人外周血CD4+、CD8+T细胞在体外培养条件下,加入雷公藤甲素处理,然后用3′末端脱氧核苷酸基(TdT)介导的dUTP缺口末端标记(TUNEL)检测凋亡细胞。结果 雷公藤甲素处理后CD4+和CD8+T细胞凋亡率比对照组显著增加(P<0.01),但CD4+、CD8+T细胞凋亡率则无显著差别(P>0.05)。结论雷公藤甲素能诱导活化的CD4+、CD8+T细胞凋亡。
ObjectiveTo investigate the effects of triptolide on apoptosis of CD4 + and CD8 + T lymphocytes. MethodsPurified CD4 + and CD8 + T cells were cultured in the presence or absence of triptolide for 12 hours.The apoptotic cells were detected by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling(TUNEL).ResultsThe apoptotic rates of CD4 + and CD8 + T lymphocytes treated with triptolide were significantly higher than that treated with ConA alone ( P <0 01),but the apoptotic rates between CD4 + and CD8 + T cells had no difference ( P >0.05).ConclusionThe immunosupression roles of triptolide are associated with nonselective induction of apoptosis of activated CD4 + and CD8 + T cells.
