摘要
目的 :观察血管紧张素 (Ang )对培养大鼠胸主动脉血管平滑肌细胞 (VSMC)增殖的作用及 AT1受体拮抗剂氯沙坦和钙通道阻滞剂硝苯地平对增殖的影响。方法 :幼龄 Wistar大鼠 (4周龄 ) 12只 ,分成对照组、Ang 组、氯沙坦加 Ang 组及硝苯地平加 Ang 组 ,采用组织块贴壁法培养胸主动脉平滑肌细胞 ,分别测定 3H-胸腺嘧啶 (3H- Td R)、3H-亮氨酸 (3H- L eu)的掺入量和细胞周期中各期细胞构成比。结果 :Ang (0 .1μmol/ L)作用 2 4h能使 VSMC的 3H- Td R与 3H- L eu的掺入量比对照组 (1%胎牛血清 )增多 ,且 S期和 G2 / M期细胞增多 ,细胞增殖指数增大。与 Ang 组相比 ,Ang 加氯沙坦 (10 μm ol/ L)组和 Ang 加硝苯地平 (10 μmol/ L)组 ,3H-Td R、3H- L eu的掺入量减少 ,S期细胞构成比和细胞增殖指数减少。结论 :Ang 对 VSMC有促增殖作用 。
To investigate the effects of losartan and nifedipine on cell proliferation induced by Ang Ⅱ in cultured rat thoracic vascular smooth muscle cell(VSMC).Method:12 young rat (4 week old) were divided into control group,AngⅡ group,losartan and AngⅡ group,nifedipine and AngⅡ group.VSMC from thoracic aorta were isolated by outgrowth of the explant method. 3H TdR and 3H Leu incorporation and cell cycle distribution were measured.Result:24 h exposure to Ang Ⅱ( 0.1 μmol/L) caused an increase in 3H TdR and 3H Leu incorporation,cell mitogenic activity also increased. The proliferation was inhibited by losartan (10 μmol/L) and nifedipine (10 μmol/L). Pretreatment with losartan and nifedipine reduced percentages of S period cells and cells mitogenic activity induced by Ang Ⅱ. Conclusion:Losartan and nifedipine antagonized the proliferation of VSMC induced by Ang Ⅱ.
