摘要
目的观察不同浓度阿霉素对大肠癌sw480细胞端粒酶活性的影响,探讨不同阿霉素作用浓度与端粒酶活性之间的关系,进而探讨阿霉素通过稳定G-四联体达到抑制肿瘤细胞增殖的可能性。方法培养sw480细胞,取对数生长期sw480细胞制成不同浓度细胞悬液,通过MTT法测定不同浓度阿霉素对sw480细胞的增殖抑制率;通过PCR-TRAT-Elisa检测不同浓度阿霉素作用下大肠癌sw480细胞的端粒酶活性。结果 MTT结果显示,阿霉素抑制sw480细胞增殖具有浓度和时间依赖性;随着阿霉素处理作用时间延长,实验组细胞端粒酶活性逐渐降低,72h后达到最低。结论阿霉素抑制sw480细胞端粒酶活性具有明显的时间和浓度依赖性,其机制可能与阿霉素稳定G四联体有关。
Objective To observe the effects of adramycin on telomerase activity of sw480 ceels,and to evaluate the relationship of the different concentration and activity of telomerase of sw480 cells,and to evaluate the possibility that adramycin restrain the proliferation of sw480 cells.Methods Cultured the sw480 cells,The effective concentration of adramycin in the experiments was determined by using methylthiazolyl tetrazolium(MTT) assay of sw480 cells,The changes of telomerase activity in sw480 cells was detected by PCR-TRAT-Elisa.Results The result of MTT declare that the effective concentration of adramycin in the subsequent experiments was determined as 5μmol/L;The activitys of telomerase of sw480 droped as the time of experiment,being the lowest at 72hours group.Conclusion Adramycin lead to inhibition of telomerase activity in sw480 cells,and the leading is determined by concentration of adramycin an the experimental time.The poosibility mechanism is adramycin's inducing telomere to form G-quadruplex structure.
