异香豆素对老年痴呆小鼠β淀粉样蛋白的影响

摘要 目的建立老年痴呆(AD)小鼠模型,探讨异香豆素对AD小鼠的治疗作用及其机制。方法将50只昆明小鼠分为:对照组、模型对照组、低剂量异香豆素治疗组、中剂量异香豆素治疗组和高剂量异香豆素治疗组,每组各10只;水迷宫实验检测各组小鼠的逃避潜伏时间和平台跨越次数;放射免疫法测定细胞培养上清Aβ含量。结果老年痴呆小鼠模型组与对照组对照比较,逃避潜伏时间出现显著性的延长(P<0.01),平台跨越次数显著性的减少(P<0.01)。异香豆素治疗后老年痴呆小鼠的逃避潜伏时间和平台跨越次数得到显著性的改善(P<0.05)。对照组、模型对照组、低剂量异香豆素治疗组、中剂量异香豆素治疗组、高剂量异香豆素治疗组脑组织Aβ的表达水平分别为(1.01±0.44)pg/ml(、12.89±3.21)pg/ml(、10.45±2.06)pg/ml、(7.18±1.92)pg/ml、(4.07±1.24)pg/ml,各组之间具有统计学差异(P<0.01)。结论异香豆素具有抗AD小鼠痴呆的活性,减少AD小鼠脑组织Aβ的生成是其主要的作用机制。
