缺血预处理对大鼠脑缺血再灌注损伤神经细胞的保护作用 被引量:3

摘要 目的探讨大鼠局灶性脑缺血预处理对脑缺血再灌注损伤后神经元的保护作用。方法健康雄性SD大鼠60只,随机分为3组:假手术组、大脑中动脉缺血再灌注(MCAO)组、预处理(BIP)组,每组按照再灌注后12 h、1、2、3 d四个时间点平均分为4个亚组,制备缺血预处理模型,分别用流式细胞术和ELISA法观察脑缺血预处理对缺血再灌注大鼠缺血半暗带神经细胞凋亡率及血清神经元特异性烯醇化酶(NSE)含量的影响。结果大鼠脑缺血再灌注后12 h,MCAO组细胞凋亡发生率及血清中NSE的含量较假手术组显著增加(P<0.01),1 d时达到高峰,以后时间点逐渐下降,但仍高于假手术组(P<0.01);BIP组各个时间点神经元凋亡发生率及血清NSE较MCAO组显著降低(P<0.05,P<0.01)。结论大鼠局灶性脑缺血预处理对脑缺血再灌注神经元损伤有保护作用。 Objective To investigate the neuron protective effect of focal cerebral ischemia preconditioning on cerebral ischemia/reperfusion(I/R) injury in rats.Methods 60 male SD rats were randomly divided into:sham-operation(SO),middle cerebral artery occlusion(MCAO) and brain ischemia preconditioning(BIP) groups.Rats in each group was further divided into 4 subgroups according to 12 h,1,2 d and 3 d after I/R.BIP models were made to measure the apoptosis rate of neuron by flow cytometry in penumbra and serum concentration of neuron-specific enolase(NSE)by ELISA after I/R.Results The rate of apoptosis neuron and NSE of rats in MCAO increased markedly at 12 h after reperfusion than that of the SO group(P0.01),and reached the peak at 1 d,then decreased continuously,but still higher than that of SO group(P0.01).BIP could decrease the rate of apoptosis neuron and NSE(P0.05,P0.01).Conclusions Focal cerebral ischemia preconditioning in rats could have a protection on neurons after I/R injury.
