摘要
选用嫁接成年树的龙眼成熟枝梢作为外植体 ,经常规消毒后 ,接种于附加不同质量浓度的激素 BA、IAA和 GA3 的 MS、N6、 1/ 2 MS、White等基本培养基上 ,诱导茎尖萌芽 .结果表明 ,MS、N6培养基适合于龙眼茎尖芽的诱导 ,最佳的激素组合为 0 .3mg·L-1BA+0 .2 mg· L-1IAA+0 .5mg· L-1GA3 .将诱导的芽转移到 MS+0 .3mg· L-1BA+0 .2 mg· L-1IAA培养基上 ,30 - 50 d内可增殖 1- 3个幼芽 ,并正常抽枝 ,形成小苗 .将小苗转入 1/ 2 MS+0 .1mg·L-1BA+0 .5mg· L-1IBA培养基 ,3- 4周后可生根 。
Mature shoots from grafted adult trees of longan were sterilized and cut into 0.5 cm shoot tips which were used as explants and inoculated onto basal media such as MS, N 6, 1/2MS and White supplemented with various mass concentrations of BA, IAA, and GA 3, in order to induce regeneration. The results showed that MS and N 6 were suitable for inducing buds from longan shoot tips and the optimal hormone combination was 0.3 mg·L -1 BA + 0.2 mg·L -1 IAA + 0.5 mg·L -1 GA 3. In the subsequent subculture, the induced buds were transferred to the medium MS + 0.3 mg·L -1 BA + 0.2 mg· -1 IAA, and 1-3 new buds were regenerated from each one in 30-50 days, and then developed into plantlets, which rooted on the medium 1/2MS + 0.1 mg·L -1 BA + 0.5 mg·L -1 IBA for 3-4 weeks and grew into normal plants.
