摘要
目的:研究不同潮气量机械通气大鼠血清对血管内皮细胞钙激活钾通道(KCa通道)的影响。方法:健康雄性SD大鼠随机分为A组(麻醉后不做任何处)和B、C组(麻醉后行气管切开插管并分别给予不同潮气量(VT)的机械通气4 h。B组VT 10 mL/kg,C组40 mL/kg),通气结束后取各组大鼠血清,分别刺激已培养好的人脐静脉内皮细胞(ECV304)30 min,利用细胞贴附式膜片钳观察KCa通道的变化。结果:B、C组内皮细胞KCa通道开放率明显上升,且以C组最为明显。结论:不同VT机械通气大鼠血清对ECV304 KCa通道的开放、关闭具有一定的影响。
Objective: To investigate the changes of Ca2 + -activated K+ channel(KCa channel) in vascular endothelial cell stmulated by serum of rats after mechanical ventilation with different tidal volumes(VT). Methods: Healthy male Sprague-Dawlay mrs were randomly divided into Croup A(no treatment after anesthesia), B and C( tracheotorny under anesthesia for ventilation with different VT for 4 hours) (VT of 10 ml/kg in the Group B and 40 mL/kg in the Group C). The serum of each rat after ventilaton was added to endothelial cell line ECV304 medium for 30 minutes, and the electric currents in the single KCa channel of the cells were recorded using patch-clamp technique with cell-attached configura- tion. Results: The open probability of the KCa channels in the Group B and C was significantly upgraded, especially in the Group C. Conclusion: The serum of rats after ventilation with different VT produces different effect to the KCa channel of ECV304.
