摘要
目的检测TBX3在非小细胞肺癌(NSCLC)组织中的表达,观察下调TBX3对肺癌细胞增殖和侵袭能力的影响。方法采用免疫组化方法检测93例NSCLC组织中TBX3蛋白的表达情况。应用siRNA干扰肺癌细胞系A549和H157内TBX3基因的表达,观察其对细胞增殖和侵袭能力的影响。结果在93例NSCLC中,71例存在TBX3蛋白过表达(76.34%),其阳性表达与肿瘤的p-TNM分期和淋巴结转移相关。转染TBX3特异性siRNA后,肺癌细胞的增殖与侵袭能力均受到显著抑制。结论 TBX3在NSCLC中呈过表达,并与肺癌分期和转移显著相关,干扰TBX3表达可明显抑制肺癌细胞的增殖与侵袭能力。
Objective To investigate the expression pattern of TBX3 in non-small cell lung cancer(NSCLC) and the effect of TBX3 knockdown on cell proliferation and invasion.Methods Immunohistochemistry was performed in 93 NSCLC tissues.Colony formation assay and transwell assay were performed in lung cancer cells after TBX3 siRNA treatment to observe its effect on cell proliferation and invasion.Results TBX3 was over-expressed in 71 of 93 cases of NSCLC.The positivity was correlated with histology,TNM stage and nodal metastasis.Knockdown of TBX3 by siRNA inhibited tumor cell proliferation and invasion.Conclusion TBX3 was over-expressed in NSCLC and correlated with lung cancer staging and metastasis.Affect the expression of TBX3 can significantly inhibit the proliferation and invasion capability of lung cancer cells.
